Is there a recommended approach to validate a FHIR message for quality and context after mapping from V2 to FHIR or is this a manual exercise completed from a clinical/domain perspective based on sample message transforms?

You'd probably want to do normal FHIR conformance stuff: define profiles for the FHIR Message and have it validated by a validator.
