I recently read a FHIR Profiles that was developed on Resources from FHIR STU 3, but the profile said it was dependent on the C-CDA on FHIR US Realm Header Profile which is for the Composition Resource from FHIR STU 1. Is this allowed/valid?

No. Profiles can only constrain profiles from the same release of FHIR. (On the other hand if it's just documentation saying "informed by/aligned with" in a hand-wavey way, that's fine.
