Is it possible to create a profile on specimen to change the type of subject to a reference to FamilyMemberHistory?

The solution there would be to have the specimen point to a Patient record and then have a link on FamilyMemberHistory to the "Patient" that represents the relative. It wouldn't make sense to have a Specimen of a a FamilyMemberHistory - the history entry doesn't represent the family member.

There's a standard extension that allows FamiliyMemberHistory to point to a Patient record for the family member
