If i want to create an extension which can be used on every element of a specific resource, how can i define this context. e.g. element/Observation would only allow the Extension directly on Observation, but not on Observation.value.

If you define it on Element, it can be used anywhere. We don't have an ability to say "everywhere, but only within a given resource type". What's the use-case for everywhere, but only in one resource type?
